摘要:
通过优化提取工艺提高新疆紫草中萘醌类成分的提取率,并研究其抗炎活性。采用 L9(34)正交设计,以左旋紫草素含量作为指标,考察药材的粒径、料液比、提取时间的影响,采用高效液相色谱法测定样品中左旋紫草素含量。以脂多糖诱导细胞炎症和二甲苯致小鼠耳廓肿胀模型考察提取物的药理作用;采用SYBYL-X软件将左旋紫草素与相关炎症蛋白进行分子对接,以初步探讨其作用机制。优选的提取工艺为:采用最粗粉第1次以1∶8的料液比提取2?h,第2次以1∶6的料液比提取1?h;细胞和动物实验结果显示其具有一定的抗炎作用,且具有剂量依赖性;分子对接结果发现左旋紫草素与环氧化酶-2具有良好的亲和能力,显示其抗炎机制可能与抑制环氧化酶-2活性有关。

Abstract:
The extraction rate of naphthoquinone components in Arnebiae euchroma was improved by optimizing the extraction technology, and the anti-inflammatory activity was studied. Using L9(34) orthogonal design, the effects of powder size, solid-liquid ratio and extraction time on the content of shikonin were investigated. The content of shikonin in samples was determined by high performance liquid chromatography. The pharmacological effects of the extracts were investigated in the model of cell inflammation induced by lipopolysaccharide and ear swelling induced by xylene in mice. SYBYL-X software was used to interconnect shikonin with related inflammatory proteins to preliminarily explore the mechanism of action. The preferred extraction technology is as follows: the coarsest powder is extracted for 2 h at 1∶8 of solid-liquid ratio for the first time, and for 1 h at 1∶6 of solid-liquid ratio for the second time. The results of cell and animal experiments showed that the shikonin has anti-inflammatory effects and its activity is dose-dependent. Molecular docking results showed that the shikonin has a good affinity with cyclooxygenase-2, suggesting that its anti-inflammatory mechanism might be related to the inhibition of cyclooxygenase-2 activity.
