摘要:
研究了枸杞子水提物的提取工艺及对酪氨酸酶活性的影响.通过正交实验优化枸杞子水提物的提取工艺,研究了料液比、提取时间、提取次数对枸杞子水提物中多糖质量分数的影响.采用蒽酮硫酸法比色测定多糖质量分数,并用体外酪氨酸酶氧化法测定枸杞子水提物对酪氨酸酶活性的影响.枸杞子水提物的最佳提取工艺为料液比120,提取时间30 min,提取次数3次,在此条件下测定枸杞子水提物的多糖质量分数为28.43%.枸杞子水提物对酪氨酸酶具有明显抑制作用,其半数抑制质量浓度IC50为0.73 mg/mL,阳性对照品熊果苷IC50为0.28 mg/mL.说明枸杞子水提物对酪氨酸酶的抑制作用与熊果苷相当.
Abstract:
The extraction technology of the water extracts from Lycium barbarum fruit was studied and the effect on tyrosinase activity was investigated.The materialliquid ratio,extraction time,extraction times were studied as 3 factors through orthogonal experiments.The content of Lycium barbarum polysaccharides was determined by phenolsulphoacid method,and the effect on tyrosinase activity was measured by the rate of oxidation of Ltyrosine.The optimum technological parameters were as follows:materialliquid ratio l20,extraction time 30 min,and extraction for 3 times.The extraction rate of Lycium barbarum polysaccharides reached 28.43% at the conditions of optimum extraction.The water extracts of Lycium barbarum fruit inhibited the activity of tyrosinase extremely,the IC50 is 0.73 mg/mL,while the IC50 of the known tyrosinase inhibitor arbutin is 0.28 mg/mL.The results showed that the water extracts from Lycium barbarum fruit express the same inhibitory effect on tyrosinase activity as arbutin.
